The new rail link, owned by Transport Scotland, is part of a key investment programme in Scotland's rail networks.
Running between Newcraighall in Midlothian to Tweedbank in the Scottish Borders, it is likely to create a substantial number of railway jobs.
The Minister for Transport, Infrastructure and Climate Change, Stewart Stevenson said the rail link would encourage economic growth across southern Scotland as well as increasing business development and housing opportunities.
It would also reduce traffic congestion and carbon emissions, he added.
Scotland's rail network currently has around 340 railway stations and 3,000 kilometres of track. Over 81 million passenger journeys are made on the network each year.
